Elpošanas ķēdes D - laktātdehidrogenāzes loma Zymomonas mobilis ndh-negatīvajā mutantā

Abstract
Darbā konstruēts baktērijas Zymomonas mobilis negatīvais mutants Zm6-ndh-ldh, un pētīta šī celma aerobā augšana un elpošana.Darba mērķis bija pētīt D - laktāta dehidrogenāzes funkcijas šīs baktērijas elektronu transporta ķēdē. D - laktātdehidrogenāzes inaktivēšana pasliktina dubultmutanta aerobo augšanu, salīdzinot ar Zm6-ndh. Zm6-ndh-ldh inaktivētā elpošanas ķēdes laktātdehidrogenāze abos pārējos celmos darbojas laktāta patēriņa virzienā, par ko liecina laktāta uzkrāšanās dubultmutanta kultivēšanas vidē. Celmam Zm6-ndh-ldh piemīt lēnāks aerobās augšanas ātrums un nedaudz zemāks biomasas iznākums nekā celmam Zm6-ndh. Secināts, ka lēnā elpošanas ķēdes darbība, izmantojot D-laktātu kā elektronu avotu, varētu būt viens no faktoriem, kurš nosaka aerobās augšanas stimulāciju celmā Zm6-ndh.
A NAD(P)H dehydrogenase and D – lactate dehydrogenase negative mutant Zm6–ndh - ldh of the bacteria Zymomonas mobilis was constructed. Its growth and respiration was studied. The aim was to study the D – lactate dehydrogenase functions in Z. mobilis electron transport chain. It is concluded that the lack of dehydrogenase decreases the aerobic growth of mutant Zm6–ndh – ldh, relative to Zm6 – ndh. The inactivated dehydrogenase in mutant strain consumes lactate, as seen by accumulation of lactate in Zm6–ndh – ldh culture. There is lower rate of aerobic growth and a little lower biomass yield in Zm6–ndh – ldh in comparison with Zm6 – ndh. The slow respiration with D - lactate could be one of the most important factors in Zm6 – ndh respiratory chain, stimulating its aerobic growth.
